摘 要:本文主要介紹了牛乳中假單胞菌屬的鑒定和PCR快速檢測。牛乳在儲存過程中,大多數是嗜冷菌,主要是假單胞菌屬和一些桿菌屬等,會產生耐受巴氏消毒甚至UHT處理的蛋白酶和脂肪酶,分解牛乳中的蛋白質和脂肪。分解產物將導致乳制品風味改變,品質降低,保質期縮短,對產品的形象和消費者的健康都造成了最大的危害。為了探索對這一問題的有效解決方法,本文對低溫貯藏原料乳中的嗜冷菌進行了研究。通過生理生化試驗,初步確定其所在的菌屬,利用通用PCR通用引物,提取假單胞菌屬的基因組DNA進行PCR快速檢測,最后進行測序比對確定其具體菌屬。

PCR For Rapid detection of Pseudomonas in milk
Abstract: This article is about PCR For Rapid detection of Pseudomonas in milk. Microorganism contamination of dairy products has been a key factor in intense market competition. When lac vaccinum is in reserve, most psychrophilic bacteria, mainly Pseudomonas and Xanthomonas, produce proteinase and lipase which tolerate pasteurization even UHT curing, reducing protein and grease. Cleavage products will change special flavor, deteriorate quality, shorten deadline, heavily influence the quality of dairy products, which will damage the products image and consumer health. In order to research the method to solve the problems, the paper studied the psychrophilic bacteria in the raw milk preserved in low temperature. This study discusses the PCR detection of Pseudomonas bacteria , and the using the bacteria common PCR primer, collected genome DNA for PCR rapid test, tested and contrasted sequence ,the confirmed the bacterium.
